The team Physical chemistry of Metals in Biology from the Laboratory of Chemistry and Biology of Metals develops the Mössbauer spectroscopy platform dedicated to iron proteins and their synthetic analogs.
Mössbauer spectroscopy is a magnetic technique that allows the study of ca forty nuclei among which iron, which makes it especially attractive for biological applications. M¨ssbauer spectroscopy detects all the iron contained in a sample whatever its chemical form or its spin state and detects exclusively iron. |
ApparatusLCBM Mössbauer platform is the only one in France dedicated to biological materials. It includes three spectrometers operating at temperatures from ca 2 K to 300 K in the pressence of magnetic fields from a few milliteslas up to 8 teslas, applied either parallel or perpendicular to the γ-ray beam. Applications Mössbauer spectroscopy allows one to characterize the active sites of iron proteins and to study their functioning. A detailed description of the electronic structure of iron centers can be reached through experiments under applied magnetic fields. Mechanistic studies of iron enzymes can be done by coupling Mössbauer spectroscopy to the rapid-mix freeze-quench technique. Usual studies are performed on isolated proteins, but experiments can be realized on cells or cells components so as to analyze global behaviors in in vivo conditions. |
